Description

The Melanocortin-3 receptor (MC3R) helps regulate energy homeostasis and mice lacking MC3R have increased fat mass and reduced lean mass. Therefore, agonists that selectively activate MC3R might have beneficial effects related to weight gain and glucose metabolism. This assay consists of a 96-well plate coated with both MC3R and Secretory Alkaline Phosphatase (SEAP) reporter constructs. Cells grown on the STEP complex will express MC3R at the cell surface. Binding of agonists to MC3R initiates a signaling cascade resulting in expression of SEAP. SEAP activity is measured following addition of luminescence-based alkaline phosphatase substrate provided in the kit.
